UJI AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL KULIT BUAH LAMTORO (Leucaena Leucacephala) TERHADAP BAKTERI Propionibacterium acnes



Acne is an inflammation that is accompanied by blockage of the ducts of the
skin's oil glands (sebum) and hair (pilosebaceous ducts). Propionibacterium acnes
is one of the causes of acne. Lamtoro (Leucaena leucacephala) is known to contain
compounds that are thought to have antibacterial potential. This study aims to
determine the antibacterial activity and concentration of ethanol extract of lamtoro
fruit peel which is effective in inhibiting the growth of Propionibacterium acnes and
to determine the content of active compounds in it.
Lamtoro fruit peel was extracted by maceration method using 96% ethanol
as solvent. Phytochemical identification was carried out qualitatively through a
color reaction with a reagent and then confirmed the content of chemical
compounds using Thin Layer Chromatography. Antibacterial activity was tested by
disc diffusion method on Mueller Hinton Agar (MHA) media. The bacteria used in
this study was Propionibacterium acnes. The ethanol extract of lamtoro fruit peel
was divided into 4 concentrations, namely 32.5%, 45%, 57.5%, 70% with
Clindamycin 1.7% as a positive control and Aquadest Sterile as a negative control.
Incubation was carried out at 37˚C for 24 hours, the parameter observed was the
formation of a clear zone around the disc.
The results of the extraction of the skin of the lamtoro fruit obtained the
extract yield of 22.87%. The results of the identification of chemical compounds
proved to contain flavonoid compounds, alkaloids and saponins. Ethanol extract
can inhibit bacterial growth and obtain the diameter of the inhibition zone at
concentrations of 32.5%, 45%, 57.5%, and 70% of 10.1 mm with an average SD ±
0.80, 10.9 mm ± 1.17, 12 .3 mm ± 0.15, and 12.9 mm ± 0.20. the most effective
concentration is at a concentration of 70%. Data analysis shows that the data is
normally distributed and homogeneous with a value of sig> 0.05.
