UJI AKTIVITAS ANTIBAKTERI EKSTRAK DAUN SINGKIL (Premna corymbosa ) TERHADAP BAKTERI Salmonella typhi SECARA IN VITRO
Singkil leaves (Premna corymbosa) contain secondary metabolite
compounds in the form of flavonoids, saponins, tannins and triterpenoids /
steroids, which can inhibit the growth of Salmonella typhi bacteria. The purpose
of this study was to determine the antibacterial activity and ethanolic extract of
singkil leaves which effectively inhibited Salmonella typhi bacteria in vitro.
Types of experimental research. Extraction of singkil leaves was carried
out using the maceration method with 70% ethanol solvent, then continued with
testing for phytochemical compounds by identifying flavonoids, saponins and
tannins. These phytochemical compounds were used as concentrations to be
tested for antibacterial activity against Salmonella typhi bacteria. Antibacterial
activity was carried out by the disc diffusion method with MHA media after
incubation for 24 hours at 37 ̊C and measuring the inhibitory diameter, namely
the clear part of the results, with a concentration of 50%, 60%, 70%, 80%, and
90% of the ethanol extract of the leaves. singkil, 30 µg chloramphenicol as a
positive control and DMSO as a negative control.
The results showed that the concentrations that gave antibacterial activity
were at concentrations of 70%, 80%, and 90% with inhibition zone values of 17.7
mm ± 0.88 SD, 18.4 mm ± 0.55 SD including the strong and 20 groups. 1 mm ±
1.05 SD is in the very strong group, and is proven to have very strong
antibacterial activity at a concentration of 90% because it shows the highest zone
