PENGARUH PERBANDINGAN PELARUT ETANOL 96% DAN ETIL ASETAT TERHADAP KADAR FLAVONOID TOTAL EKSTRAK DAUN TAPAK DARA (Catharantus roseus)



Tapak dara (Catharanthus roseus) is a plant that grows a lot in Indonesia. Tapak
dara contains various active chemical substances such as tannins, triterpenoids
and flavonoids. Tapak dara leaves are efficacious as a diuretic, hypotensive,
sedative, hemostatic. The purpose of this study was to determine the flavonoid
content of 96% ethanol extract and ethyl acetate extract of tapak dara leaves
using UV Vis spectrophotometry method. Tapak dara leaves were extracted by
maceration using 96% ethanol and ethyl acetate as solvents. Both extracts were
tested for their secondary metabolite content by tube test and thin layer
chromatography (TLC). Determination of total flavonoid levels using the UV-Vis
spectrophotometric method with quercetin as a reference standard at a
wavelength of 414.5 nm and the data were analyzed using the Kolmogorovsamirnov normality test and homogeneity then continued with the one-way Anova
test. The results of the phytochemical screening test carried out with the tube test
of tapak dara leaves contain flavonoids, tannins, and saponins. Thin layer
chromatographic analysis on 96% ethanol extract and ethyl acetate contains
flavonoids, namely the appearance of yellow spots with an Rf value of 0.85.
Based on the one-way ANOVA test, there was no significant difference in the
total flavonoid content of the ethanol extract 30.3 mgQE/gextract and ethyl
acetate 28.6 mgQE/gextract.
