Singkil leaves (Premna corymbosa) contain secondary metabolites in the
form of flavonoids, saponins, tannins, and triterpenoids / steroids, capable of
inhibiting the growth of Staphylococcus aureus. The purpose of this study was to
determine the antibacterial activity of the ethanolic extract of singkil leaf and to
determine the concentration of the ethanolic extract of singkil leaf extract which
was effective against Staphylococcus aureus in vitro.
Singkil leaf extraction was carried out using the maceration method with
70% ethanol solvent, then continued with phytochemical compound testing by
identifying flavonoids, saponins, and tannins by means of phytochemical
screening. The next step is to test the antibacterial activity using the disc method,
the bacteria used are Staphylococcus aureus, the positive control used
Amoxicilin 10 g disk and the negative control DMSO 10% and the media used
was MHA (Mueller Hinton Agar) then the concentration of the extract used was
50% , 60%, 70%, 80%, and 90%, then incubated at 37ºC for 1x24 hours. The
clear zone obtained was measured and analyzed by the Kruskal-Wallis test.
The results showed that the inhibition zones against Staphylococcus
aureus bacteria at concentrations of 60, 70, 80, and 90% were 8.7; 18.8; 19.0;
and 22.9 mm. The most effective concentration was 90%, while the 50%
concentration had no antibacterial activity. The analysis of the data shown is not
normal and homogeneous, then the results of the Kruskal-Wallis test are that
there is a significant difference between the concentrations of 50% and 90%.
