PENENTUAN KONSENTRASI HAMBAT MINIMUM (KHM) DAN KONSENTRASI BUNUH MINIMUM (KBM) FRAKSI ETIL ASETAT EKSTRAK ETANOL DAUN AFRIKA (Vernonia amygdalina Del.) TERHADAP Propionibacterium acnes



The bacteria that causes infection in acne is Propionibacterium acnes.
African leaves (Vernonia amygdalina Del.) in Indonesia are empirically widely
used by the community by drinking boiled water. The purpose of this study was to
determine the antibacterial activity of the ethyl acetate fraction of the ethanolic
extract of African leaves against Propionibacterium acnes bacteria and the
chemical compounds contained in the ethyl acetate fraction of African leaves.
African leaves were extracted by maceration method using 96% ethanol
as solvent. African leaf ethanol extract was fractionated by liquid-liquid extraction
method using chloroform and ethyl acetate as solvents. The ethyl acetate fraction
obtained was identified qualitatively by phytochemical and TLC test using silica
gel plate GF254 then made in series with concentrations of 100, 250, 500, 750
and 1000 ppm, then tested against Propionibacterium acnes using liquid dilution
method to determine MIC and solid dilution method to determine MBC. The
positive control used was erythromycin 1000 ppm and the negative control used
was DMSO 10%. In this study, all activity were incubated at 370C. The MIC
parameters observed were the absorbance value of the test solution before and
after incubation and the presence or absence of bacterial growth in the MBC test.
The results of the ethyl acetate fraction of African leaves contain
flavonoid compounds, tannins, saponins, alkaloids, and steroids or terpenoids.
The concentration of 750ppm was set as the Minimum Inhibitory Concentration
while the Minimum Bactericidal Concentration could not be determined because
there was still bacterial colony growth in the test.
